The rail line between Portadown and Belfast Great Victoria Street will be closed this coming Sunday, September 11.
Bus substitution services will be in place on the day between Belfast and Portadown, serving Lisburn, Lurgan and Moira.
Enterprise services will also be closed with a bus service operating between Belfast and Newry.
Translink’s Helen Halliday said: “This closure will allow us to carry out waterproofing at Stockmans Lane bridge and drainage works at a number of level crossings on the line between Lisburn and Portadown.
“Passengers wishing to travel on this date should arrive at their departure point in good time and allow extra time to reach their destination. Valid rail tickets will also be accepted on bus services operating on Lisburn Road in Belfast.
“We have placed passenger information notices at all impacted locations. Local residents have been advised of the works and we would like to thank passengers for their patience.”
